On Tuesday, JP Morgan highlighted its survey related to&nbsp;Apellis Pharmaceuticals,&nbsp;Inc.&#8217;s&nbsp;(NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/APLS#NASDAQ">APLS</a>) Syfovre and Astellas Pharma Inc&#8217;s (OTC:<a class="ticker" href="https://www.benzinga.com/stock/ALPMF#OTC">ALPMF</a>) (OTC:<a class="ticker" href="https://www.benzinga.com/stock/ALPMY#OTC">ALPMY</a>) <a href="https://www.benzinga.com/general/biotech" target="_blank" rel="noreferrer noopener">Izervay</a>.
On average, surveyed physicians treated around 388 geographic atrophy patients and most of them had used Syfovre and Astellas&#8217; Izervay (~80%).
JP Morgan analyst Anupam Rama writes that the survey results point to steadier growth for Syfovre/branded geographic atrophy products over the next 12 months versus an inflection-type growth.

Survey data indicates that perceptions of Syfovre and Izervay have remained largely unchanged before and after the Izervay CRL and label update. Syfovre is seen as slightly more effective, while Izervay has a safety advantage.
The geographic atrophy market is expected to respond well to promotional efforts. Over time, Apellis&#8217; strategy—including direct-to-consumer marketing, targeting younger physicians, increasing prescribing depth and emphasizing its unique dosing—should drive growth.
The company generated Syfovre sales of $167.8 million and $611.9 million for the fourth quarter and full year 2024, respectively.
It delivered approximately 94,000 Syfovre doses to physician practices in the fourth quarter, including approximately 89,000 commercial vials and 4,600 samples. More than 510,000 Syfovre injections, including clinical trials, are estimated to have been administered since <a href="https://www.benzinga.com/pressreleases/25/02/g44038782/apellis-pharmaceuticals-reports-fourth-quarter-and-full-year-2024-financial-results" target="_blank" rel="noreferrer noopener">launch through December 2024</a>.
Taking a cautious view, the analyst projects a longer timeline for Syfovre to reach peak sales, valuing it along with Empaveli (for PNH) at around the mid-$30s per share—still leaving room for growth from current levels.
JP Morgan maintains an Overweight rating while lowering the <a target="_blank" href="https://www.benzinga.com/quote/APLS/analyst-ratings" rel="noreferrer noopener">price target to $45 from $54</a>, based on extending time to peak and peak sales for Syfovre and adjustments to expenses.

The results showed a sustained overall survival (OS) and progression-free survival (PFS) benefit consistent with the findings of the primary analysis after an additional 12 months of follow-up (median follow-up of 29.1 months).

<li>Results showed enfortumab vedotin plus pembrolizumab reduced the risk of death by 49% versus chemotherapy.</li>
<li>The median OS was 33.8 months for the combination versus 15.9 months for chemotherapy.</li>
<li>The OS benefit was observed in all prespecified subgroups, including cisplatin eligible and ineligible subgroups.</li>
<li>Enfortumab vedotin plus pembrolizumab also reduced the risk of disease progression or death by 52% versus chemotherapy.</li>
<li>The median PFS was 12.5 months for the combination versus 6.3 months for chemotherapy.</li>
<li>The safety profile was consistent with previous findings and no new safety concerns were identified.</li>
</ul>
In addition to longer follow-up data, an exploratory analysis evaluating treatment outcomes and safety profile in patients with confirmed complete response (cCR) will also be presented.
<ul>
<li>Among patients evaluable for response, a confirmed objective response rate (cORR) was 67.5% for enfortumab vedotin plus pembrolizumab compared to 44.2% for chemotherapy.</li>
<li>Median duration of response (DOR) was 23.3 months for the combination and 7.0 months for chemotherapy.</li>
<li>A cCR was achieved in 30.4% of patients treated with enfortumab vedotin plus pembrolizumab and 14.5% of patients treated with chemotherapy.</li>
<li>The median duration of cCR was not reached for the combination and 15.2 months for chemotherapy.</li>
<li>In patients with cCR, grade ≥3 treatment-related adverse events occurred in 61.7% of patients in the enfortumab vedotin plus pembrolizumab arm compared to 71.9% in the chemotherapy arm. There were no treatment-related deaths in the cCR subgroup.</li>

Analyst Andy Hsieh writes that investing in Elevation Oncology offers a favorable risk-to-reward ratio, especially at its current valuation. He has initiated coverage with <a href="https://www.benzinga.com/quote/ELEV/analyst-ratings" target="_blank" rel="noreferrer noopener">an Outperform rating</a>.
The company provides exposure to the fast-growing market of antibody-drug conjugates and focuses on a scientifically and regulatory-validated target. The ADC field has gained significant interest, with claudin 18.2 emerging as a promising target.

In August, Elevation Oncology reported initial data from the dose escalation portion of the ongoing <a target="_blank" href="https://www.benzinga.com/general/biotech/24/08/40196487/why-is-penny-stock-elevation-oncology-trading-lower-on-tuesday" rel="noreferrer noopener">Phase 1 trial of EO-3021</a>.
As of the data cutoff date of&nbsp;June 10, 32 patients had been treated. EO-3021 was observed to be generally well-tolerated. No Grade 4 or 5 treatment-related adverse events were reported, and less than 10% of patients discontinued EO-3021 due to adverse events.
Hsieh estimates the total addressable market for frontline CLDN18.2-positive gastric and GEJ cancer in five major regions at about $6.6 billion, with $1.8 billion in the U.S. alone.
For second-line pancreatic cancer, which has limited treatment options, the TAM is projected at $1.8 billion across the U.S. and Europe, including $1.3 billion in the U.S. The analysts believe even a small market share in these areas could significantly boost the company&#8217;s current valuation.
Recently, Elevation Oncology nominated EO-1022 as its HER3 ADC development candidate. EO-1022 is progressing through preclinical development, and Elevation Oncology expects to file an investigational new drug <a target="_blank" href="https://www.benzinga.com/pressreleases/24/12/n42458079/elevation-oncology-expands-pipeline-with-nomination-of-eo-1022-a-her3-adc-for-the-treatment-of-her" rel="noreferrer noopener">application in 2026</a>.

Elevation Oncology: Promising Cancer Therapies Earn Analyst Praise

On Monday, Sangamo Therapeutics, Inc.&nbsp;(NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/SGMO#NASDAQ">SGMO</a>) regained the development and commercialization rights to giroctocogene fitelparvovec, an investigational gene therapy product candidate for moderately severe to severe hemophilia A that it has co-developed with and licensed to Pfizer Inc (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/PFE#NYSE">PFE</a>).
Pfizer decided to terminate the global collaboration and license agreement <a target="_blank" href="https://www.benzinga.com/pressreleases/24/12/b42733065/sangamo-therapeutics-to-regain-full-rights-to-hemophilia-a-gene-therapy-program-following-pfizers-" rel="noreferrer noopener">between the parties</a>. Sangamo intends to explore all options to advance the program, including seeking a potential new collaboration partner.

In July 2024, Pfizer announced topline results from the Phase 3 AFFINE trial of giroctocogene fitelparvovec, which demonstrated that the trial met the primary and key secondary objectives of superiority <a target="_blank" href="https://www.benzinga.com/general/biotech/24/07/39937203/sangamo-stock-spikes-as-pfizer-partnership-meets-goal-in-late-stage-study" rel="noreferrer noopener">compared to prophylaxis</a>.
&#8220;&#8230;we believe it is well positioned for regulatory submissions and potential commercialization,&#8221; said Sandy Macrae, Chief Executive Officer of Sangamo Therapeutics. &#8220;While we were surprised and extremely disappointed by Pfizer&#8217;s decision to end our collaboration so close to the anticipated BLA and MAA submissions&#8230;&#8221;
The collaboration and license agreement with Pfizer will terminate effective April 21, 2025. All trial participants will continue to be monitored as planned during the transition period.
Sangamo believes that its recently announced partnerships with Roche Holdings AG&#8217;s (OTC:<a class="ticker" href="https://www.benzinga.com/stock/RHHBY#OTC">RHHBY</a>) Genentech and Astellas Pharma Inc (OTC:<a class="ticker" href="https://www.benzinga.com/stock/ALPMF#OTC">ALPMF</a>) (OTC:<a class="ticker" href="https://www.benzinga.com/stock/ALPMY#OTC">ALPMY</a>) and advanced business development discussions for its Fabry gene therapy program will allow it to chart a path forward for its neurology genomic medicine pipeline.
The company prepares to initiate the expected enrollment of patients in the Phase 1/2 study of ST-503 for idiopathic small fiber neuropathy in mid-2025 and file an anticipated Clinical Trial Authorisation submission for the prion disease program in Q4 2025, each subject to securing additional funding.

Sangamo Therapeutics Stock Plunges As Pfizer Terminates Hemophilia Gene Therapy Pact

William Blair has initiated coverage on Apellis Pharmaceuticals, Inc. (NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/APLS#NASDAQ">APLS</a>) and notes that Apellis was the first company to successfully target complement C3, which has led to the approval of two therapies: Empaveli (systemic pegcetacoplan) for paroxysmal nocturnal hemoglobinuria and Syfovre (intravitreal pegcetacoplan) for geographic atrophy secondary to age-related macular degeneration.
William Blair writes that although Empaveli has established a solid position in the PNH market, short-term challenges are anticipated due to the emergence of a new competitor, Novartis AG&#8217;s (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/NVS#NYSE">NVS</a>) Fabhalta (iptacopan).

However, Empaveli has a more significant market opportunity in C3 glomerulopathy (C3G) and immune-complex membranoproliferative glomerulonephritis (IC-MPGN), especially following the strong top-line results from the Phase 3 VALIANT study.
William Blair projects that it could receive approval by late 2025, with peak sales estimates for C3G and IC-MPGN reaching $700 million by 2031.
The analyst has initiated the coverage with an Outperform rating and estimates a fair value for <a target="_blank" href="https://www.benzinga.com/quote/APLS/analyst-ratings" rel="noreferrer noopener">shares of $50</a>.
Syfovre received approval in February 2023, while Astellas Pharma Inc&#8217;s (OTC:<a class="ticker" href="https://www.benzinga.com/stock/ALPMF#OTC">ALPMF</a>) (OTC:<a class="ticker" href="https://www.benzinga.com/stock/ALPMY#OTC">ALPMY</a>) drug was approved in August 2023. According to the analyst, Syfovre presents a more attractive efficacy profile and a more advantageous label, permitting monthly or bi-monthly dosing and treatment beyond 12 months (although the label for Izervay may be updated to include these options in November). 
Nevertheless, the analyst notes that the market is sufficiently large to accommodate multiple blockbuster drugs.
Thus, even though Syfovre has a competitive advantage over Izervay, William Blair believes both medications can thrive in light of the extensive market, significant unmet needs, and the absence of alternative treatments.
With an assumption of only 30% peak market penetration among patients treated by injecting physicians and a consistent rise in referral rates without an increase in diagnosis rates, the analyst projects U.S. peak sales for Syfovre to reach $2.0 billion by 2030.

The company completed an internal analysis of efficacy and safety data from Phase 1 monotherapy and Phase 1b combination studies of NUV-868.
Following this analysis, Nuvation Bio decided not to initiate a Phase 2 study of NUV-868 as a monotherapy or combined with olaparib or enzalutamide in the advanced solid tumor indications that were part of the Phase 1 and Phase 1b study designs.
The company is evaluating the next steps for the NUV-868 program, including further development in combination with approved products for indications in which BD2-selective BET inhibitors may improve patient outcomes.
&#8220;As we focus on our late-stage pipeline and prepare to potentially bring taletrectinib to patients in the U.S. in 2025, we have decided not to initiate a Phase 2 study of NUV-868 in the solid tumor indications studied to date. Two years back, Nuvation Bio discontinued the clinical development of NUV-422 after an internal risk-benefit analysis factoring in feedback from the FDA in a partial clinical hold letter for monotherapy Phase 1/2 study and clinical hold letters for its <a target="_blank" href="https://www.benzinga.com/general/biotech/22/08/28290264/after-fda-hold-nuvation-bio-stops-nuv-422-cancer-development-program" rel="noreferrer noopener">combination Phase 1b/2 studies</a>.

Zolbetuximab, a first-in-class claudin (CLDN) 18.2-targeted monoclonal antibody, is recommended in combination with fluoropyrimidine- and platinum-containing chemotherapy for the first-line treatment of adult patients with locally advanced unresectable or metastatic human epidermal growth factor receptor 2 (HER2)-negative gastric or gastroesophageal junction (GEJ) adenocarcinoma whose tumors are CLDN18.2 positive.1 If approved, zolbetuximab would become the first and only CLDN18.2-targeted therapy available for patients in the European Union.
