Subjects receiving the Natrunix ON+5FU+LV regimen also had about a 33% reduction in hospitalization (80 days versus 120 days).
Subjects receiving the Natrunix combination also reported a 22% reduction in fatigue, 32% improved appetite, and 41% reduction in pain.
The incidence of severe diarrhea was twofold (9% versus 19%) lower during the 24-week treatment regimen for patients receiving the Natrunix + ON+5FU+LV combination compared to placebo + ON+5FU+LV.
Overall Survival (OS) is one of the secondary endpoints for the Phase 2 study. The data highlights the observation that no subjects in the placebo ON+5FU+LV group (n=32) survived for longer than 330 days, whereas eight subjects in the Natrunix ON+5FU+LV arm (n=33) were still alive as of day 330.
Considering the small sample size, the borderline statistically significant p-value of p = 0.096 suggests prolonged survival for subjects receiving the Natrunix regimen.

When administered immediately prior to artery-opening procedures, Hutrukin may reduce the inflammatory injuries associated with reperfusion. Twenty-two leading cancer centers across the United States have been involved in the Phase I/II study. Natrunix blocks a key cause of inflammation involved in pain and joint destruction in rheumatoid arthritis (RA).

The primary endpoint of the Phase II is the American College of Rheumatology (ACR)&nbsp;20%&nbsp;Response (ACR 20) rate at 12 weeks. Secondary and exploratory endpoints include ACR 50, ACR 70, numerical rating scale (NRS) for pain score, inflamed and painful joint counts, Health Assessment Questionnaire Disability Index (HAQ-DI), Routine Assessment of Patient Index Data 3 (RAPID-3), Clinical Disease Activity Index (CDAI) and safety. These endpoints reflect the expectation that Natrunix may provide needed improvement with respect to pain, tenderness, mobility of joints and quality of life. Approximately 210 subjects will be enrolled into three different arms in the study. All subjects will receive methotrexate (MTX) and be randomized to receive either one of two doses of Natrunix or placebo.
Natrunix is a monoclonal antibody indistinguishable from a naturally occurring antibody from a human donor. Natrunix binds and neutralizes the action of one of the most potent inflammation-causing substances known&mdash;interleukin-1. For decades, interleukin-1 was seen as the key target for drugs to treat RA. However, to date, drugs that block interleukin-1 have not lived up to expectations as therapies for RA. XBiotech believes it has solved this puzzle.
Interleukin-1 actually describes two separate and distinct molecules&mdash;IL-1a and IL-1b&mdash;that are produced at different times and places in the body. XBiotech is the first to develop candidate therapies&mdash;like Natrunix&mdash;that directly and specifically neutralize IL-1a. In recent years, world-class research has shown that in many cases IL-1a may be the crucial target for blocking disease-causing activities of the interleukin-1 inflammatory pathway. XBiotech Commences Tender Offer To Purchase Up To $80M Worth Of Its Shares At Price Not Greater Than $4.00 Nor Less Than $3.80 Per Share

&nbsp;XBiotech Inc. (NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/XBIT#NASDAQ">XBIT</a>) has enrolled the first subject in a randomized, open-label, placebo-controlled dose escalation clinical study to evaluate safety and pharmacokinetics of Hutrukin. 
&nbsp;
Hutrukin is being developed as a novel treatment to reduce brain injury following ischemic stroke. There are more than 10 million ischemic strokes each year&mdash;the leading cause of death and morbidity in the world today. An ischemic stroke occurs when a blood clot forms in a blood vessel that supplies blood flow to brain. Blockage of blood flow to the brain results in brain injury, loss of brain function or death.
Emergency use of &quot;clot-busting&quot; drugs or mechanical catheters to re-open arteries after a stroke is associated with a phenomenon known as&nbsp;reperfusion injury. Reperfusion injury is where the return of blood supply after removing the clot from the artery results in increased brain injury or death. Reperfusion injury is believed to be the result of a massive inflammatory response caused by blood cells as they flow back into the region of the brain that was deprived of blood supply (and oxygen).
Hutrukin therapy is intended to be provided immediately prior to the clot-busting procedure, where the drug may reduce inflammatory injury associated with reperfusion. There is currently no drug known to be effective to reduce reperfusion injury. Hutrukin could, therefore, potentially represent a significant advance in the management of stroke patients.
The current Phase I study will evaluate three dose levels of Hutrukin for safety and will measure the corresponding blood levels of Hutrukin with increasing doses. Findings will guide dosing and safety expectations for the next phase of studies.
