On Monday, Silence Therapeutics plc (NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/SLN#NASDAQ">SLN</a>) presented end-of-treatment data from its Phase 2 ALPACAR-360 study of zerlasiran in atherosclerotic cardiovascular disease (ASCVD) patients with high lipoprotein(a) [Lp(a)] <a target="_blank" href="https://www.benzinga.com/pressreleases/24/11/b42046710/silence-therapeutics-presents-late-breaking-phase-2-zerlasiran-data-at-2024-american-heart-associa" rel="noreferrer noopener">levels (≥125 nmol/L)</a>.
These data were presented at the American Heart Association (AHA) Scientific Sessions.
Results showed that zerlasiran (300 mg every 16 weeks, 300 mg every 24 weeks or 450 mg every 24 weeks) produced greater than 80% mean time-averaged placebo-adjusted reductions from baseline in Lp(a) concentrations over 36 weeks.
This is the first study to report time-averaged Lp(a) analyses, which more accurately evaluate the effects of treatment over time, including intervals between doses.
Maximum Lp(a) reductions exceeded 90%. At the final visit, 60 weeks following initial drug administration, reductions in Lp(a) persisted, and no safety concerns emerged with infrequent dosing.
William Blair notes that while Silence's data shows the drug effectively lowers Lp(a) levels, the Q24W dosing schedule may not be competitive. This is due to a decline in Lp(a) reduction between doses.
This dosing schedule could be an option for maintenance after stable Lp(a) reductions are reached. However, the analyst expects more frequent dosing, possibly every three months, to be needed. This may reduce zerlasiran's competitive edge compared to Amgen Inc.'s (NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/AMGN#NASDAQ">AMGN</a>) olpasiran.
The analyst writes, "Silence continues to execute on both the zerlasiran and divesiran lead clinical programs. While data for zerlasiran presented at AHA show that zerlasiran is an efficacious Lp(a) lowering agent over 48 weeks with Q24W dosing, we are incrementally negative on zerlasiran's competitive profile given the waning of Lp(a) lowering between the infrequent doses, which has not been demonstrated with olpasiran at a once-quarterly dosing frequency."
Price Action: SLN stock is down 33.80% at $7.20 at the last check on Tuesday.

Silence Therapeutics' Zerlasiran Has Competitive Concerns: Analyst

Analysts expect Target Corporation (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/TGT#NYSE">TGT</a>) to report third-quarter revenue of $25.9 billion when the company reports third-quarter financial results on Wednesday, Nov. 20 <a href="https://www.benzinga.com/earnings">before the market opens.</a>
 That&#8217;s up from $25.4 billion in last year's third quarter, according to data <a href="https://www.benzinga.com/pro/?utm_source=BZprocontent111924&amp;t=be2bearmibe3na1">from Benzinga Pro.</a>
The Minneapolis-based company has beaten analyst estimates for revenue in six of the last eight quarters and seven of the last 10 quarters overall.
Analysts expect Target to report third-quarter earnings per share of $2.30 — up from $2.10 in last year's third quarter. The company has beaten analyst estimates in six of the last seven quarters and six of the last 10 quarters overall.
<a href="https://www.benzinga.com/news/earnings/24/08/40482203/target-q2-earnings-revenue-and-profit-beat-digital-sales-boost-annual-profit-outlook-hiked">Guidance from Target </a>calls for third-quarter comparable sales to be up 0% to 2% year-over-year. The company also guided for third-quarter earnings per share to be in the range of $2.10 to $2.40.
Target raised its full-year guidance after second-quarter results with earnings per share of $9.00 to $9.70 now expected, up from a prior range of $8.60 to $9.60.

<h3 class="wp-block-heading">Key Items to Watch</h3>
Target visitor traffic was up 1% year-over-year, per data from Placer.ai. It outpaced retail rival Walmart Inc (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/WMT#NYSE">WMT</a>), which had 0.9% visitor growth year-over-year.
The Placer.ai <a href="https://www.placer.ai/blog/superstores-and-wholesale-clubs-ahead-of-the-holidays">report</a> also sheds some light on upcoming trends to watch for Target and how the company's guidance could factor into the holiday season. Target saw the biggest weekly holiday visits boost of the tracked retail chains, with foot traffic up 87.3% the week before Christmas compared to the company's weekly average. 
Walmart, by comparison, saw a 39.6% increase in its weekly average visitors for the week before Christmas last year.
"Digging deeper into historical data suggests that it is Target that is primed to experience the busiest holiday season of the analyzed chains," Placer.ai said.
Target also saw the biggest spike of the tracked retailers for the weekly visitor growth for the week of Turkey Wednesday and Black Friday, with visits up 18.9% compared to the company's weekly average.
"With recent visits to Target on par with — or slightly above — last year's levels, the retail giant is likely poised to win the holidays once again."
Along with guidance and commentary on the holiday shopping season, another area to watch in Target's report will be same-day delivery services. The company reported a double-digit increase in this segment in the second quarter.
Sales by segment could also be an area to watch with Target saying apparel and discretionary sales were strong in the second quarter.
Target's quarterly results come one day after Walmart <a href="https://www.benzinga.com/24/11/42060801/walmart-tops-q3-estimates-ecommerce-growth-shines-with-27-lifts-full-year-forecast">reported third-quarter results</a>. Walmart reported revenue and earnings per share that came in ahead of Street consensus estimates. The retailers also raised its guidance.
The strong quarterly report from Walmart could be a positive for Target heading into the earnings report as it could signal the strength of the American consumer. The key will be making sure that Walmart didn't end up taking market share away from Target as a result.
Price Action: Target stock is down 1% to $155.37 on Tuesday versus a 52-week trading range of $128.37 to $181.86. Target stock is up 8.6% year-to-date in 2024.

On Monday, Aclaris Therapeutics, Inc.&nbsp;(NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/ACRS#NASDAQ">ACRS</a>) entered into an exclusive license agreement with Biosion Inc for worldwide rights (excluding Greater China) to BSI-045B and BSI-502, a preclinical-stage, novel bispecific antibody directed against <a target="_blank" href="https://www.benzinga.com/pressreleases/24/11/g42034751/aclaris-therapeutics-announces-exclusive-global-license-agreement-with-biosion-inc-adding-potentia" rel="noreferrer noopener">both TSLP and IL4R</a>.
In a completed Phase 2a, single-arm, proof-of-concept trial in the United States in 22 patients with moderate to severe atopic dermatitis, BSI-045B was observed to have a pharmacodynamic, safety, and efficacy profile.
BSI-045B is also being advanced in multiple Phase 2 studies in China by Biosion&#8217;s regional partner, Chia Tai Tianqing Pharmaceutical Group, Co., Ltd. (CTTQ), targeting severe asthma and chronic rhinosinusitis with nasal polyps, accelerating the potential to show proof-of-concept across additional indications.
Under the terms of the agreement, Biosion shall receive over&nbsp;$40 million&nbsp;cash payment as an upfront license fee and reimbursement for certain development costs &amp; drug product materials.
Biosion shall also receive 19.9% shares of Aclaris Therapeutics.
Additional regulatory and sales milestone payments exceed&nbsp;$900 million, with tiered low-to-mid single-digit royalty payments based on a percentage of <a target="_blank" href="https://www.benzinga.com/pressreleases/24/11/n42036056/biosion-announces-exclusive-global-license-agreement-with-aclaris-therapeutics-on-two-potential-fi" rel="noreferrer noopener">annual net sales</a>.
Leerink Partners upgraded Aclaris from Market Perform to Outperform, with a price target of $7, <a target="_blank" href="https://www.benzinga.com/quote/ACRS/analyst-ratings" rel="noreferrer noopener">up from $2</a>.
The analyst writes, &#8220;Following completion of their strategic review, we expect investor focus to shift to ACRS&#8217; newly strengthened immunology-focused pipeline.&#8221;
Leerink writes that these updates mark a fresh start for Aclaris Therapeutics, with a clear strategic direction and promising catalysts in the next 6-12 months. This progress could reduce risks for BSI-045B and significantly enhance its value.
The analyst has updated the model to include BSI-045 B&#8217;s potential in atopic dermatitis (AD), estimating around $1.3 billion in revenue by 2035 with a 35% probability of success. The analyst also anticipates multiple Phase 2 trial results from China in asthma and chronic rhinosinusitis with nasal polyps, which could provide more insight into BSI-045 B&#8217;s potential.
Additionally, Leerink expects further progress in de-risking the company&#8217;s pipeline in the first half of next year.
BTIG upgrades Aclaris Therapeutics from Neutral to Buy and announces a $8 price target.

The retail investing landscape is changing as individuals are increasingly looking to take on more risk. Yoni Assia, co-founder and CEO of investing platform eToro, believes crypto is playing a big role in the <a href="https://www.benzinga.com/money/retail-investors-ve-institutional-investors">evolution of the retail investor</a>.

Older generations focused on dividend yields, the Baby Boomers started focusing on price-to-earnings multiples, Gen X began focusing more on growth, Millennials took it a step further and focused on tech stocks and now Gen Z is more focused on crypto and things like Dogecoin (CRYPTO: <a class="ticker" href="https://www.benzinga.com/quote/doge/usd">DOGE</a>) becoming the currency of Mars, he said.
EToro started the year with just 2% of its customers trading crypto and now has more than 90% of customers involved with crypto in some capacity across its platform, Assia said.
Crypto is driving that bigger tolerance for risk due to its becoming a &#8220;generational asset class&#8221; and a &#8220;gateway trade&#8221; for a lot of individual investors. Many retail investors come to the platform to trade Bitcoin (CRYPTO: <a class="ticker" href="https://www.benzinga.com/quote/btc/usd">BTC</a>), for example, and then end up exploring other investment options, he said.
Many crypto investors have a higher appetite for risk and as crypto investors cross into traditional markets, retail traders are starting to make a bigger splash. The surge in retail investors in recent years has changed market dynamics, Assia explained.

The way investors get information has also changed. In the past, a lot of individuals would read investment reports and put a lot of time into researching a potential investment, but now traders are getting information very quickly from various news outlets and from other individual investors on social media platforms like Reddit and X. If they are reading research reports, they are likely using ChatGPT or another LLM to pull the key information, he added.
When talking about retail investors versus individual investors, you often hear the terms &#8220;dumb money&#8221; and &#8220;smart money,&#8221; but &#8220;I think that&#8217;s completely wrong,&#8221; Assia said: &#8220;We see it on our platform.&#8221;
eToro allows investors to see what other investors on the platform are doing and even copy their trades. Assia told the crowd at the Benzinga conference that there are 3,200 investors on the platform that are being copied by other traders. Most of those investors consistently generate double-digit returns and often outpace the S&amp;P 500. 
&#8220;These are like hedge-fund level returns of our top retail investors on the platform. I think this narrative is coming from the people who want to manage the retail&#8217;s money and don&#8217;t want them to manage their own money,&#8221; he said. 
The playing field is already in the process of being leveled and new AI tools are only going to help accelerate retail&#8217;s ability to see consistent success in the markets, Assia said.

Crypto trader More Crypto Online hopes that Bitcoin can move forward toward $95,000 and then maybe $97,000. On the other hand, the trader <a href="https://x.com/Morecryptoonl/status/1858936812082807069">remains</a> cautious suggesting this could be a corrective range. He believes the structure lacks clear direction and momentum and is therefore very fragile.
Santiment data noted that with Bitcoin peaking to all-time highs, whale transactions and retail FOMO could make the "short-term price action unpredictable, but the long-term indicators for Bitcoin looking quite sound."
