Biogen and Idec merged in 2003, combining forces to market Biogen's multiple sclerosis drug Avonex and Idec's cancer drug Rituxan. Today, Rituxan and next-generation antibody Gazyva (oncology) and Ocrevus (multiple sclerosis) are marketed via a collaboration with Roche. Biogen markets several multiple sclerosis drugs including Plegridy, Tysabri, Tecfidera, and Vumerity. Biogen's newer products include Spinraza (SMA, with partner Ionis), Leqembi (Alzheimers, with partner Eisai), Skyclarys (Friedreich's Ataxia, Reata), Zurzuvae (postpartum depression, Sage), and Qalsody (ALS, Ionis). (NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/BIIB#NASDAQ">BIIB</a>) – announced the initiation of dosing in the global clinical study, PROMINENT. The Phase 3 study will evaluate the efficacy and safety of the investigational drug felzartamab compared to tacrolimus in adults diagnosed with primary membranous nephropathy (PMN). PROMINENT is designed to enroll approximately 180 adults with PMN and expected to readout in 2029. PMN is a severe antibody-mediated disease of the kidney that is a leading cause of nephrotic syndrome and carries a significant risk of kidney failure.
Felzartamab is an anti-CD38 antibody that has been shown in clinical studies to selectively deplete CD38+ cells, including plasma cells, the source of autoantibodies that mistakenly attack the body's own tissues in a range of immune-mediated diseases. Importantly for felzartamab, it is estimated that up to 80% of PMN patients have autoantibodies against PLA2R (aPLA2R) generated by CD38-expressing plasma cells. Currently, there are no approved treatments for PMN and the standard of care includes treatments ranging from immunosuppressants to chemotherapy.

Biogen Initiates Dosing In Global Clinical Study, PROMINENT, For Treatment Of Primary Membranous Nephropathy

These latest findings from Part C of the DEVOTE trial evaluating a higher dose regimen of nusinersen and the NURTURE trial which evaluated the approved 12 mg regimen (SPINRAZA&#xAE;) in clinically presymptomatic SMA were presented at the SMA Research &amp; Clinical Care Meeting hosted by Cure SMA in Anaheim, Calif. Biogen's applications for the higher dose regimen of nusinersen are currently under review in the U.S., Europe, Japan and other global markets. The higher dose regimen of nusinersen comprises a more rapid loading regimen – two 50 mg doses 14 days apart – and a higher maintenance regimen – 28 mg every four months.
Improvements Observed With Higher Dose Regimen of Nusinersen in Previously Treated Patient Population Detailed results from Part C of the DEVOTE study highlight the potential clinical benefits of an investigational higher dose of nusinersen in a broad range of individuals (n=38) who had been previously treated with nusinersen at the approved 12 mg dose for approximately four years (median: 3.9 years). Participants were 4 to 65 years of age and half (n=19) were ambulatory. Participants in Part C received one loading dose (50 mg) and two maintenance doses (28 mg each) of open-label higher dose nusinersen during the study period.
Most participants experienced improvements on the Hammersmith Functional Motor Scale – Expanded (HFMSE), Revised Upper Limb Module (RULM), and/or Clinical Global Impression of Change (CGI-C; assessed by investigator or caregiver) after transitioning to the higher dose regimen. These improvements were observed across phenotypes, functional status and age. For example, non-ambulatory participants improved by +2.5 (95% CI: 0.49, 4.56) on average on the HFMSE, and ambulatory participants improved by +1.1 (95% CI: -0.68, 2.89).

The safety profile of the higher dose regimen of nusinersen is broadly consistent with the known safety profile of 12 mg SPINRAZA. In the DEVOTE study overall, reported adverse events (AEs) included pneumonia, respiratory failure, pyrexia, COVID, upper respiratory tract infection, procedural pain and procedural headache. In Part C (n=40), AEs were reported in 37/40 participants, the majority of which were mild or moderate in severity. Serious AEs were reported by six participants (15%), none of which were considered by the investigator to be related to study treatment or administration.
Final NURTURE Data Redefine Expectations for Early Treatment Final data from the eight-year, open-label NURTURE study highlight the impact of early intervention with 12 mg SPINRAZA in clinically presymptomatic infants with SMA.
At the study conclusion, all children who participated in NURTURE (n=25) were alive and experienced continued clinical benefits over the course of the study. No participants required permanent ventilation, and the majority (20 of 25 participants) went without any ventilatory support throughout the study. Ninety-two percent of participants achieved the ability to walk independently, many within normal developmental timeframes. Participants with elevated levels of neurofilament light chain (NfL) at baseline experienced rapid and sustained reductions in NfL after initiation of nusinersen, reinforcing the potential utility of NfL as an objective biomarker of disease activity and treatment response in SMA.
Nusinersen was generally well tolerated with no new safety concerns identified with eight years of follow-up. All participants had at least one AE, the majority of which were mild to moderate in severity; no AEs led to treatment discontinuation or study withdrawal.

An interim analysis of the phase one study in participants with SMA who were previously treated with Novartis AG&#8217;s (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/NVS#NYSE">NVS</a>) gene therapy, Zolgensma (onasemnogene abeparvovec), was conducted to inform the decision on whether to move salanersen forward <a href="https://www.benzinga.com/pressreleases/25/06/g46091748/biogen-to-advance-investigational-spinal-muscular-atrophy-asset-to-registrational-studies-based-on" target="_blank" rel="noreferrer noopener">into registrational studies</a>.
Both dose levels tested, 40 mg and 80 mg, given once a year, were generally well-tolerated and led to substantial slowing of neurodegeneration, as shown by reductions in neurofilament.
Exploratory clinical outcome data show clinically meaningful improvements in function and attainment of new World Health Organization (WHO) milestones over one year.
Interim results are from part B (n=24) in individuals who received either 40 mg or 80 mg salanersen once a year.
In participants with elevated baseline concentrations of neurofilament light chain (NfL), indicating ongoing neurodegeneration, initiation of salanersen led to mean reductions in NfL of 70% at six months, which were sustained through the one-year dosing interval.
Exploratory clinical outcome data were evaluated for the subgroup of participants with at least one year of follow-up during the interim analysis (n=8 participants aged 2-12 who received 40 mg of salanersen). Half (four out of eight) of these participants achieved new WHO motor milestones they could not previously achieve on their own or required assistance, such as walking, crawling, standing or sitting.
Furthermore, these participants experienced clinically meaningful improvements in motor function from baseline to one year, including a 3.3-point (SD 4.46) mean improvement from baseline on the Hammersmith Functional Motor Scale – Expanded (HFMSE) and a 5.3 point (SD 4.75) improvement on the Revised Upper Limb Module (RULM).
Biogen is currently engaging with global health authorities regarding the design of the phase 3 studies.
Earlier in June, Biogen initiated dosing in the BRAVE phase 3 study to evaluate the efficacy and safety of omaveloxolone in children with Friedreich ataxia (FA) between <a href="https://www.benzinga.com/pressreleases/25/06/g45992415/biogen-initiates-phase-3-pediatric-study-of-omaveloxolone-for-the-treatment-of-friedreich-ataxia" target="_blank" rel="noreferrer noopener">the ages of 2 to &lt;16</a>.
Both non-ambulatory and ambulatory participants may qualify for the study.
Currently, omaveloxolone is commercialized under the brand name Skyclarys in over 40 countries, including the U.S. and the European Union, and is the only approved product for FA in adults and adolescents aged 16 years and older.

<li>A higher proportion of those receiving DZP (19.2%) versus SOC alone (8.4%) also achieved disease remission at Week 48.</li>

Participants from the PHOENYCS GO study will continue to be followed in a long-term open-label study. A second Phase 3 trial of dapirolizumab pegol, PHOENYCS FLY, is ongoing.
In May, Biogen and City Therapeutics, Inc., a privately held biopharmaceutical company, announced a strategic collaboration to develop <a target="_blank" href="https://www.benzinga.com/pressreleases/25/05/g45614561/biogen-and-city-therapeutics-announce-strategic-research-collaboration-to-develop-select-novel-rna" rel="noreferrer noopener">select novel RNAi therapies</a>.
The collaboration will initially focus on a single target that mediates key central nervous system diseases. Biogen will be responsible for IND-enabling studies and global clinical development along with any regulatory submissions and all activities related to commercialization.
City Therapeutics will receive $46 million in payments, including a $16 million upfront payment and an investment of $30 million in exchange for a City Therapeutics convertible note, representing a minority equity interest in the company if converted.
Biogen will record the upfront payment as an Acquired In-Process Research and Development expense in the second quarter of 2025.
Should the initial program achieve certain development and commercial milestones, City Therapeutics can receive up to approximately $1 billion in potential milestone payments plus tiered royalties in the high single-digit to low double-digit range based on net sales.
Biogen will have the option to select one additional target in the collaboration, subject to an additional payment and availability of the target.
